A recently granted patent (Publication Number: US11884962B2) discloses a method for generating nucleic acid barcode molecules. The method involves providing a plurality of nucleic acid molecules coupled to a support, coupling additional nucleic acid molecules to assemble a variety of nucleic acid barcode molecules, and digesting off-target products with an exonuclease. The target products contain an exonuclease-resistant bond, with less than 50% of these bonds being digested by the exonuclease.

The patent claims also detail the use of specific exonucleases such as Exo1 exonuclease, Exo111 nuclease, or heat-labile DNAse in the method. Additionally, the method specifies the absence of reducing agents like dithiothreitol during the digestion process. The temperature range for the digestion step is set between 37 to 65 degrees Celsius for a duration of 15 minutes to 1 hour. Furthermore, the support for the nucleic acid molecules can be a bead, specifically a gel bead, with the digestion process not affecting the integrity of the bead. The method also involves coupling capture sequences to the nucleic acid molecules during the assembly of the barcode molecules.
