Each well in the array extends into a planar surface of the substrate, with a fluid pathway passing across the array of wells. The genetic material is isolated with a population of probe-coupled particles at the array of wells, achieving a capture efficiency of over 80%. Process reagents are delivered into the fluid pathway, a layer of oil is distributed to prevent transfer between wells, heat is transmitted to process the genetic material, and the contents of each well are optically interrogated.
Furthermore, the method includes capturing the probe-coupled particles in single-particle format at a large number of wells, delivering process reagents using a pump, and distributing the layer of oil through pressure generation. Heat transmission involves probe hybridization and thermocycling, with a reflective heating surface reflecting light back into the wells. The substrate also features a fluid reservoir sealed by a lid with grooves for displacing fluid phases. The method allows for real-time fluorescence imaging, redistribution of contents exceeding well capacity, quantitative analysis of genetic material, and generation of a genotyping library. The fluid pathway includes inlet and outlet channels, and the genetic material can be derived from single cell lysate, with a lysis buffer delivered as part of the process reagents.
