The granted patent US12054766B2 outlines a method for incubating and analyzing a liquid sample using a specialized device. The process involves depositing a liquid sample between a substrate's surface, which is coated with a water-insoluble pressure-sensitive adhesive, and a polymeric cover film. This cover film is a composite material that includes a polymer and a tackifier. By applying external pressure, discrete regions of the cover film are brought into contact with the adhesive, creating multiple closed compartments that contain the liquid sample. The method facilitates cell division within these compartments and allows for a most probable number analysis of microorganisms, such as E. coli or S. aureus, without the need for serial dilutions or risk of cross-contamination.

The device can contain between 100 to 300 microcompartments, each designed to hold microvolumes of liquid. The method also includes the option of using a nutrient growth medium, which may be selective and contain indicator substances. Additionally, the incubation can be performed in an anaerobic environment for anaerobic microorganisms, with temperature conditions ranging from 25°C to 45°C. The design of the device allows for effective sealing of the compartments, ensuring that the liquid remains contained during the analysis process. This innovative approach enhances the efficiency and accuracy of microbial analysis in laboratory settings.
