Rapid Micro Biosystems has been granted a patent for a general-purpose microbiological growth media that supports the growth of microorganisms on membranes. The media, which does not contain added sodium, includes casein digest, soybean digest, animal tissue digest, yeast extract, dextrose, a phosphate buffer, hemin, and L-cystine. This media is capable of supporting the growth of anaerobes, molds, injured spores, and general aerobic bacteria more effectively than other media. A recently granted patent (Publication Number: US11732237B2) discloses a composition for culturing cells. The composition consists of casein digest, soybean digest, animal tissue digest, yeast extract, dextrose, a phosphate buffer, hemin, and L-cystine. Notably, the composition does not contain added sodium.

The composition can exist in different forms depending on the temperature. At 22°C, it is in a solid state, specifically in the form of a powder. The composition comprises varying amounts of the different components, excluding the phosphate buffer, ranging from 1 g/kg to 500 g/kg for casein digest, soybean digest, animal tissue digest, yeast extract, and dextrose.

The composition also includes a quantity of phosphate buffer that provides a buffer capacity of 0.1 mmol/(pH unit) to 100 mmol/(pH unit) when dissolved in an aqueous medium.

At a different temperature, the composition can be in a liquid or gel form. In this case, it may contain purified water and has a pH of 7.3±0.5.

Importantly, the composition does not contain tris(hydroxymethyl)aminomethane.

Additionally, the composition can include a disinfectant neutralizer, such as histidine, thiosulfate, polysorbate 80, and/or lecithin.

The patent also describes a method of culturing cells using this composition. The method involves contacting a population of cells with the composition under conditions that support their growth. The cells can be placed on one side of a membrane, with the other side of the membrane in contact with the composition.

Overall, this patent presents a composition and method for culturing cells that does not contain added sodium and can exist in different forms depending on the temperature. The composition includes various components, excluding the phosphate buffer, in specific ranges, and can be used for cell culture applications.
