The image size is then magnified from the end of the optical fiber to a first dichroic filter in a de-multiplexing imaging array. The fluorescent light is reflected between a series of dichroic filters and micro-mirrors, allowing for collimation and re-imaging of the light. Different wavelength ranges of the fluorescent light are then band-passed at each dichroic filter to de-multiplex the wavelength spectrum. The fluorescent light in each wavelength range is detected to count the number of particles tagged with each fluorochrome in the sample fluid.

The method also includes additional steps to enhance the accuracy of the analysis. Before magnifying the image, laser light launched from the optical fiber is blocked to prevent interference with the detection of fluorescent light. The fluorescent light launched from the optical fiber is collimated and focused onto the first dichroic filter. Prior to detection, the different wavelength ranges of the fluorescent light are focused into a plurality of light detectors.

In order to efficiently direct the fluorescent light to a compact detection module, the method involves receiving the fluorescent light generated by fluorochromes excited by laser light with a collection lens. The light is then converted from a first numeric aperture to a second numeric aperture matching that of the optical fiber. The fluorescent light is coupled into the opposite end of the optical fiber for flexible direction.

Additionally, the method includes a second set of steps for analyzing a second wavelength range of the fluorescent light. The fluorescent light is split into the first and second wavelength ranges, and the image size is magnified to a second dichroic filter in a second de-multiplexing imaging array. The second wavelength range is reflected between dichroic filters and micro-mirrors, and different wavelength ranges are band-passed to de-multiplex the spectrum. The fluorescent light in each wavelength range is then detected to count the number of particles tagged with each fluorochrome.

The patent also describes the use of a telescopic effect created by the focal length of the micro-mirrors and the distance of separation between the dichroic filters and micro-mirrors. This effect allows for collimation and re-imaging of the fluorescent light on the dichroic filters.
